Calu-3细胞

Calu-3是经常应用于癌症研究药物开发的人类肺癌细胞系,最初于1975年由纪念斯隆-凯特琳癌症中心科研人员分离自一名25岁高加索男性肺癌患者胸腔积液。

特征

ERBB2基因在Calu-3细胞中过度表达,并在侵袭性癌症的扩散中发挥作用。

Calu-3细胞以粘附的单层形式生长及增殖,而且对CK-7英语Keratin 7封闭蛋白英语Occludin和E-钙粘蛋白阳性反应,同时具有大量的囊肿性纤维化跨膜调节器英语Cystic fibrosis transmembrane conductance regulator[1],并且因过度表达ERBB2基因而令ErbB2/Her2被激活[2]

科研用途

Calu-3细胞可以在临床前研究中充当呼吸系统模型,并且因其对异物的反应能力强而成为吸气及肺损伤模型[3]。多份研究指出Calu-3细胞通常用作体外体内抗肺癌药物开发的模型[3]、显示摄入低分子物质的能力而有助于肺部药物的输送的研究[4]、研究肺上皮细胞分泌离子的过程[1]。Calu-3细胞可以应用于重点在肺细胞屏障完整性和表面蛋白表达的高速化合物筛选英语High-throughput screening[5]
